RNA Purifier: Preparative HPLC system for batch purification of RNA

with ternary pump, UV detection and thermal control

The therapeutic potential of mRNA is extensively discussed in times of search for vaccines in the research of COVID-19. The idea is to provide the human body with mRNA, encoding therapeutic proteins, which the immune system uses to fight the virus.1
RNA can be produced and applied much easier compared to the full-length therapeutic proteins it is encoding for. In order to generate pure mRNA to administer, it was found that purification by chromatographic means is highly important.2
Key for RNA chromatography is the HPLC system layout. It is highly important that the chromatography is carried out at elevated temperatures in order to denature the RNA itself and hence simplify its structural diversity and yield a successful purification. Eleveated temperature in combination with a binary gradient between acetonitrile and water as done in the Reverse Phase Chromatography Method, will effectively seperate and also inactivate and unfold RNase.3,4
The dedicated RNA Purifier is an especially designed system for RNA purification. It consists of a ternary low pressure system pump, which not only gives you a reliable formation of binary gradient between water including 1.0 M TEAA (pH 7) and acetonitrile, but also allows the direct sample application of large volume sample onto the column by the system pump via its separate sample inlet port. The flow rate range of this pump lies between 10 to 80 ml/min. Sample Injection via a sample- or Variloop is possible with the integrated injection valve. After the injection valve solvent as well as sample will be heated by an eluent heater and then applied onto the column, which additionally is heated by a heating sleeve to ensure perfect thermal purification conditions. Product identification is realized by the variable single wavelength detector at 260 nm or the wavelength of your choosing. After purification fractionation is carried out by the fractionation valve.

Automated sample preparation system TrayMix™ S4

CHARACTERISTICS:

Operation: Automatic

Applications: Laboratory, for research, diagnostic, cytogenetic

Sample type: Washing, for hybridization

Configuration: Benchtop

Preparation format: for microscope slides, biochip

DESCRIPTION:

Fully Automated Hybridization Station The automated hybridization and wash station TrayMix™ S4 is a micro-mixer using a patented active mixing by Chaotic Advection technology which belongs to BioTray®. Best results are obtained thanks to an homogeneous dispersion of molecules over the whole hybridization area (21 x 60 mm). The patented Active Mixing by Chaotic Advection system achieves a significant reduction of hybridization times with the use of lower samples quantity (5 pmoles) ; a higher specificity can be reached (high reduction of background noise, increased fluorescent signal intensities). The patented Active Mixing by Chaotic Advection system eliminates artefacts such as dead zones or bubbles usually observed with other systems so that you can succeed in your hybridization and wash experiments from the start.

PCR preparation system Xdrop™

CHARACTERISTICS:

Operation: Automatic

Applications: Laboratory, for PCR, NGS

Sample type: for DNA

Preparation format: droplets

Configuration: Benchtop

Preparation format: High-throughput, with touchscreen, high-speed

DESCRIPTION:

The Xdrop instrument is a droplet generator that can produce droplets with use of specific microfluidics double emulsion and single emulsion cartridges. It is operated from the touch screen and droplet generation is fully automated. The choice of cartridge determines the size and properties of the droplet. In this way samples can be partitioned in droplets as standalone compartments for reactions. A white gasket (rubber cover) provides an air-tight seal between the instrument and cartridge during operation. Specifications Width: – 25 cm – 9,8 inches Height: – 25 cm – 9,8 inches Length: – 48 cm – 18,9 inches Weight: – 17 kg – 37,5 lbs. Voltage requirements: – 110 V-240 V Cartridges Samplix has developed advanced microfluidics easy-to-use Xdrop cartridges for droplet generation in the Xdrop instrument. Double emulsions cartridge [called dPCR cartridge] This cartridge produces double emulsion droplets, which can be used as compartments for high-throughput screening and identification of the target. Encapsulate millions of molecules or small cells (up to 6µmdiameter) with Xdrop. Run 8 samples simultaneously or individually 40 minutes of droplet generation time Produce 4-10 million droplets per lane Contamination-free workflow Single emulsions cartridge [Called dMDA cartridge] This cartridge produces single emulsion droplets, which can be used as compartments for single molecule amplification. Run 8 samples simultaneously or individually 40 seconds of droplet generation time Produce > 50,000 droplets per sample and per run Contmination-free workflow.
